The peak and trough were observed at 390 and 420 nm, respectively, for various substrate concentrations. The peak-to-trough absorbance differences were plotted against the respective substrate concentrations for determining the Kd value using the equation Aobs = Amax (([S]+[Et]+KD) - (([S]+[Et]+KD)2 - (4[S][Et])0.5)/2[Et].
