(A) To confirm the disruption of MRS3/4, genomic DNA from the wild-type strain and the mrs3/4 mutant was digested with Xho1/SacI and hybridized with the indicated probes. (B) Southern blot analysis indicated genomic deletion of MRS3/4. Two independent mrs3/4 mutants (#1 and #2) were constructed, and #1 was used throughout the study.
