Fig. 3. Effect of PTPRB on the mitochondrial content of brown adipocytes. (A) NADH dehydrogenase 1 (ND1) was used to measure the mitochondrial DNA (mtDNA). Genomic and mtDNA were isolated from brown adipocytes using DNA Mini Kits. TATA box binding protein (TBP) primers were used to assess the nuclear DNA. (B) The amount of mitochondria was assessed after ectopic expression of the wild-type PTPRB or PTPRB mutant. Cells were stained with MitoTracker fluorescence (Invitrogen) for visualization of mitochondria using confocal microscopy. MitoTracker is identified by the red color, and DAPI-stained nuclei are blue.
